Clorsulon Structure

Synonymes : 4-Amino-6-(trichloroethenyl)-1,3-benzenedisulfonamide, MK-401, Clorsulonum, 4-Amino-6-(trichlorovinyl)-m-benzenedisulfonamide

Clorsulon is a benzenesulfonamide derivative characterized by a trichloroethenyl group and two sulfonamide moieties. This unique structure enables it to inhibit key enzymes in the glycolytic pathway of parasitic flukes, particularly phosphoglycerate kinase and phosphoglyceromutase, disrupting energy production in susceptible organisms.

First developed for veterinary medicine, Clorsulon is primarily used to treat liver fluke infections in cattle and sheep. It is available as a high-purity active pharmaceutical ingredient (API) with a minimum assay of 98.0%, meeting stringent quality standards for pharmaceutical and research applications.

In addition to its veterinary use, Clorsulon serves as a reference standard and is investigated for its potential in other therapeutic areas. Its well-defined mechanism and favorable safety profile make it a valuable compound for both veterinary and pharmaceutical research.

Structure chimique et propriétés

Clorsulon belongs to the sulfonamide class and features a benzene ring substituted with an amino group, a trichloroethenyl group, and two sulfonamide groups. It appears as a white to off-white crystalline powder, slightly soluble in water but soluble in organic solvents like methanol and ethanol. The compound is stable under normal storage conditions and exhibits good thermal stability.

Mécanisme d'action

  • Enzyme inhibition: Inhibits phosphoglycerate kinase and phosphoglyceromutase, disrupting glycolysis in flukes.
  • Energy depletion: Leads to ATP depletion and subsequent death of the parasite.
  • Selective toxicity: High efficacy against Fasciola species with low toxicity to mammalian hosts.
  • Synergistic potential: Often used in combination with other anthelmintics for broader spectrum activity.
