Cefuroxime axetil is a second-generation cephalosporin antibiotic and the orally active prodrug of cefuroxime. Its structure features a 7-aminocephalosporanic acid core with a methoxyimino-furyl side chain at C-7 and an acetoxyethyl ester at C-3. This ester modification increases lipophilicity and allows efficient absorption from the gastrointestinal tract, after which nonspecific esterases in the intestinal mucosa and blood rapidly hydrolyze the ester to release the active cefuroxime.
It is supplied as a white to off-white crystalline powder, practically insoluble in water but freely soluble in acetone and methanol. The compound is manufactured under cGMP conditions and is available in different polymorphic forms, with Form I and Form II being the most common.
