Aminophylline Structure

Synonyms: Theophylline ethylenediamine, Aminophyllinum, Euphyllin, Phyllocontin, 317-34-0, Theophylline compound with ethylenediamine (2:1).

Aminophylline is a pharmaceutical salt formed from theophylline and ethylenediamine in a 2:1 molar ratio. It was developed to overcome the poor water solubility of theophylline, allowing for rapid preparation of injectable and oral solutions. While theophylline itself is a naturally occurring methylxanthine found in tea leaves, aminophylline is a synthetic derivative that has been used in clinical practice for decades, primarily as a bronchodilator in respiratory care.

Chemically, aminophylline is not a single molecule but a molecular complex. The ethylenediamine component enhances aqueous solubility, while theophylline remains the active pharmacophore. This allows aminophylline to deliver theophylline more efficiently in acute settings. The compound is typically standardized to contain 84.0–86.0% theophylline and 13.5–15.0% ethylenediamine, corresponding to an assay of 98.0–102.0% on the dried basis.

As an API, aminophylline is supplied in high-purity grades suitable for formulation into injections, tablets, and oral liquids. It is also used as a reference standard and in research applications. Our material is manufactured under cGMP conditions, with full documentation to support regulatory submissions.

Function & Mechanism

  • Bronchodilation: Relaxes bronchial smooth muscle by inhibiting phosphodiesterase, increasing cyclic AMP.
  • Respiratory stimulation: Improves diaphragmatic contractility and reverses fatigue.
  • Anti-inflammatory: Reduces airway inflammation through adenosine receptor antagonism.
  • Cardiac stimulation: Positive inotropic and chronotropic effects at therapeutic doses.
  • Diuretic: Increases renal blood flow and glomerular filtration rate.

Source & Standardization

  • Source: Synthetic theophylline reacted with ethylenediamine.
  • Manufacturing: Controlled crystallization and drying to achieve consistent particle size.
  • Standardization: Assay 98.0–102.0% (HPLC, dried basis); theophylline content 84.0–86.0%.
  • Forms: White to off-white crystalline powder.
  • Applications: Injectable solutions, tablets, oral liquids, research.
