Clove Oil Structure

Synonyme: Eugenia caryophyllata oil, Clove bud oil, Caryophyllus aromaticus oil, Syzygium aromaticum oil, Oil of cloves

Clove oil is a volatile essential oil obtained from the dried flower buds, leaves, and stems of the clove tree, Syzygium aromaticum. Its characteristic aroma and biological activity stem largely from eugenol, a phenylpropanoid that makes up 70–95% of the oil. The remaining fraction contains eugenyl acetate, β-caryophyllene, and minor terpenes that contribute to its overall profile. The oil is typically produced by steam distillation, a process that preserves the delicate aromatic compounds. Clove oil is available in several grades, including USP/NF and food-grade, and is standardized to specific eugenol content to suit different applications.

In the pharmaceutical and nutraceutical industries, clove oil is valued for its antimicrobial, analgesic, and antioxidant properties. It is commonly incorporated into oral care products, topical analgesics, and digestive aids. The oil also serves as a natural preservative in cosmetics and a flavoring agent in food and beverages. Its broad spectrum of activity makes it a staple in formulators’ toolkits.

Chemical Structure & Properties

Eugenol (4-allyl-2-methoxyphenol) is the primary bioactive constituent, featuring a phenolic hydroxyl and an allyl side chain. This structure underlies its ability to disrupt microbial membranes and scavenge free radicals. Clove oil is a complex mixture; besides eugenol, it contains eugenyl acetate (up to 15%), β-caryophyllene (5–12%), and trace amounts of other sesquiterpenes. The oil is lipophilic, with a density of about 1.04 g/mL, and is sensitive to light and oxygen, which can cause discoloration and loss of potency. Proper storage under nitrogen, in airtight containers away from light, helps maintain its quality.

Mechanism of Action

  • Antimicrobial activity: Eugenol disrupts bacterial and fungal cell membranes, leading to leakage of intracellular contents and cell death.
  • Analgesic effect: It inhibits voltage-gated sodium channels and modulates TRPV1 receptors, reducing pain perception.
  • Antioxidant action: The phenolic group scavenges free radicals, protecting cells from oxidative stress.
  • Anti-inflammatory: Suppresses COX-2 and other inflammatory mediators.
